Histone proteins provide structural support for chromosomal DNA and regulate the accessibility of genes for transcription.1 Giant cell tumors of bone are locally aggressive primary bone tumors that usually occurs in the epiphysis of long bones in young adults. Giant cell tumors of bone harbor a histone protein mutation (H3.3 G34W) and show expression of the H3.3 G34W protein.1-4 Immunohistochemistry for H3.3 G34W shows a nuclear pattern of staining.
