Cystatins are endogenous inhibitors to cysteine proteinases. They inhibit In various degrees the cysteine proteinases such as the mammalian cathepsins B, H, L, and S. Cystatin A (also termed acid cysteine proteinase inhibitor, ACPI, stefin A, and epidermal SH-proteinase inhibitor), a 98 amino acid protein, is expressed in the basal epithelial cells of normal and hyperplastic prostate (as well as PIN1 and PIN2) and this expression is absent in prostate carcinoma. The utility of Cystatin A antibody is similar to that of CK 34betaE12 and CK 5 & 6 in the discrimination of prostate carcinoma vs. benign prostate. This protein is also expressed in normal squamous epithelium, follicular dendritic cells in lymphoid tissues, thymic epithelial cells, liver cells, and granulocytes.
